    You wont be able to kill the 500 enemies in a single run, you will still need around 20 to 40 more enemies after you finish the game but dont worry.

    Simply start a new game and continue on killing, this counter wont reset if you begin a new game.

    You could kill some enemies, restart the battle and do them again, the counter will go up each time, so in theory you could get the 500 in a single game if you pruposefully restart battles for this but.. its far easier to finish the game and restart the first or second battles until this pops instead.
