The boss mode characters can be bought from the collections menu for 5,000 C each. Note that in order to buy a boss character, you first need to either finish arcade mode with the regular version of that character, or buy both of their endings.

To switch to boss mode, hold down LB while selecting your character. The achievement should unlock at the start of the match.
